چکیده

High-affinity receptors for human granulocyte macrophage colony-stimulating factor (GM-CSF), interleukin-3 (IL-3). and IL-5 are composed of two distinct subunits, a and p. Each receptor has its own ligand-specific a subunit, and the three receptors share the common p subunit, BE. Using a transfectant of NIH3T3 cells expressing the high-affinity human GM-CSF receptor, monoclonal antibodies (MoAbs) against pc were generated. These MoAbs specifically bound to cells bearing pc and immunoprecipitated the pc protein of 120 Kd. Using these MoAbs, expression of pc was examined. It is known that IL-1 augments the proliferative response of a

RAPID COMMUNICATION Hematopoiesis in Mice Lacking the Entire Granulocyte-Macrophage Colony-Stimulating Factor/Interleukin-3/Interleukin-5 Functions

Interleukin-3 (IL-3). granulocyte-macrophage colony-stimulating factor (GM-CSF), and IL-5 are major hematopoietic cytokines produced by activated T cells and exhibit similar biologic activities by signaling through a common receptor subunit (pc). Mice lacking p c show a pulmonary alveolar proteinosis-like disease and reduced numbers of peripheral eosinophils, which are explained by the lack of ...

Simultaneous antagonism of interleukin-5, granulocyte-macrophage colony-stimulating factor, and interleukin-3 stimulation of human eosinophils by targetting the common cytokine binding site of their receptors.

Human interleukin-5 (IL-5), granulocyte-macrophage colony-stimulating factor (GM-CSF), and IL-3 are eosinophilopoietic cytokines implicated in allergy in general and in the inflammation of the airways specifically as seen in asthma. All 3 cytokines function through cell surface receptors that comprise a ligand-specific alpha chain and a shared subunit (beta(c)). Although binding of IL-5, GM-CSF...

The Human Granulocyte - Macrophage Colony - Stimulating Factor ( GM - CSF ) Receptor Exists as a Preformed Receptor Complex That Can Be Activated

The granulocyte-macrophage colony-stimulating factor GM-CSF. The preformed complex required only the extracellular regions of GMRa and bc , as shown by the ability of (GM-CSF) receptor is expressed on normal and malignant hematopoietic cells as well as on cells from other organs soluble bc to associate with membrane-anchored GMRa or soluble GMRa. Kinetic experiments on eosinophils and in which ...

Monoclonal antibody 7G3 recognizes the N-terminal domain of the human interleukin-3 (IL-3) receptor alpha-chain and functions as a specific IL-3 receptor antagonist.

The human interleukin-3 receptor (IL-3R) is expressed on myeloid, lymphoid, and vascular endothelial cells, where it transduces IL-3-dependent signals leading to cell activation. Although IL-3R activation may play a role in hematopoiesis and immunity, its aberrant expression or excessive stimulation may contribute to pathologic conditions such as leukemia, lymphoma, and allergic reactions. We d...

Neutrophils Activated by Granulocyte-Macrophage Colony-Stimulating Factor Express Receptors for Interleukin-3 Which Mediate Class I1 Expression

Freshly isolated peripheral blood neutrophils, unlike monocytes and eosinophils, do not bind interleukin-3 (IL-3) or respond t o IL-3. We show that neutrophils cultured for 24 hours in granulocyte-macrophage colony-stimulating factor (GM-CSF) express mRNA for the IL-3 receptor (R) a subunit, as shown by RNase protection assays, and lL-3Rn chain protein, as shown by flow cytometric analysis usin...
